A simple infection from the G-Virus caused the Nemesis and its host to mutate instead of the statis the Nemesis parasite gave the host. This is when he got half nakie and grew bigger tenticles and turned into a bigass hellspawn at the later parts of the games.
Just saying that could've happened to Thanatos-R.
But of course, someone could inject Tyrant-R to make him into that big fucker at the end of the trailer...Seeing as thats like the biggest mutation in the game so far, not including Project: Alexia's hive ass.

Of course, Thanatos is purely made with the T-Virus. If he were to be injected, he would've been an ultimate monster, because G-Virus uses deadly mutation DNA, and if mixed with T-Virus which almost uses the same procedure could involve catastrophic.
And Thanatos-R, I believe, was destroyed at the end of REO by the big explosion. The epilogue at the end informed that there were still T-Virus spreading. I'm saying in the File number 2 of REO, some scientists could've manufactered the same DNA and added some G-Virus into it.

And look on the trailer, do you see a blocked entrance of firey cars/burning deribis of buildings when that big creature who we suspect is Thanatos is?

Of course, REO covers the gaps. It also unveils what kind of monstrosities Umbrella can really create. And, you might not know this since you are into oldie RE, but REO was split up into three files. REO File 1, REO File 2 and REO File 3. The third file supposedly has five missions, so in total, REO has 15 missions together.
And yes, I did see that, but it is not the debris from REO the first.
